RainDance Expands Sequence Enrichment Solution Capabilities with 20,000 Amplicon Library and Whole Genome Amplification Protocol

In a presentation at Cambridge Healthtech Institute’s Exploring Next-Generation Sequencing conference, the company said these new enhancements represent a logical evolution of the RainDance technology platform and will add attractive benefits for the researcher investigating rare variants in large populations that cause cancer and other devastating diseases.

Expanded Primer Library Enables Broader View of Human Genome

RainDance said it has expanded the content of its DNA primer library from 4,000 amplicons to 20,000 amplicons, while maintaining the precision provided by microdroplet-based PCR. This will allow researchers to incorporate a broader range of content for their large-scale targeted resequencing studies and to broaden their view of the human genome.

“Workflow and sample requirements will not change as the content is scaled up to 20,000 amplicons,” McNary said. “Our Sequence Enrichment Solution will maintain its performance and continue to provide the highest-quality data and sample uniformity.”

The company said the expanded DNA libraries will be available in the first quarter of 2010 and can be ordered through RainDance sales channels and service providers.

Whole Genome Amplification Protocol Supports Small Sample Sequencing

RainDance also announced it is the first company to support the use of whole genome amplified DNA samples for sequence enrichment. This capability will enable researchers to conduct studies with sample quantities that were previously too small for targeted resequencing. Now scientists can perform whole genomic amplification (WGA) on rare or small samples that have very low amounts of DNA, such as those found in tumor biopsies.

“The results of our studies demonstrate that RainDance’s Sequence Enrichment Solution is fully compatible with WGA samples and delivers the same high-quality results as assays performed with unamplified data,” said McNary.

The company said its WGA protocol is available now and has been validated using kits from Qiagen, GE Healthcare, and BioHelix Corporation. The kits can be ordered from their respective manufacturers.

“Both our expanded content library and WGA protocol are exceptionally easy to use and work seamlessly with our Sequence Enrichment Solution and RDT 1000 system,” McNary said. “These new capabilities are the first of many exciting applications for microdroplet-based PCR that RainDance will introduce in the coming weeks. RainDance Technologies Announces Shipment of RDT 1000, Sequence Enrichment Solution to TGen Researchers

TGen, a nonprofit biomedical research institute based in Phoenix, Arizona, focuses on the translation of laboratory discoveries into personalized medicine, developing earlier diagnostics and better treatments for patients with many forms of cancer, neurological and cardiovascular diseases, and infectious diseases.

Dr. David Duggan, an Investigator in TGen’s Genetic Basis of Human Disease Division, said one of the initial applications of the RainDance solution would focus on resequencing regions of the human genome identified as part of genome-wide association studies.

In an announcement at the European Society of Human Genetics (ESHG) Conference 2009, the company said ATLAS Biolabs’ purchase includes the RDT 1000 system, PCR primer libraries, consumables kits, and training services.

RainDance Technologies Announces First Commercial Shipment of RDT 1000, Sequence Enrichment Solution

– Delivery to Ontario Institute for Cancer Research –

LEXINGTON, MASSACHUSETTS, U.S.A., March 26, 2009 — RainDance Technologies, Inc., a provider of innovative microdroplet-based solutions for human health and disease research, today announced the first commercial shipment of its RDT 1000 and Sequence Enrichment Solution. The shipment will be delivered to the Ontario Institute for Cancer Research in Canada (OICR), a new, innovative institute dedicated to research in the prevention, early detection, diagnosis and treatment of cancer.

OICR will use the RainDance solution – consisting of a RDT 1000, consumables kits, custom PCR primer libraries, and expert training – in its programs involving the discovery and validation of DNA sequence variants associated with cancers. The solution is designed to increase quality and efficiency of performing targeted sequencing on large sample numbers.

The RDT 1000 and Sequence Enrichment Solution utilize RainDance’s breakthrough RainStorm™ microdroplet-based technology platform. The simplicity and speed of the technology are designed to maximize the efficiency of next-generation DNA sequencing workflows. The RDT 1000 generates picoliter volume PCR reactions at the rate of 10 million discrete reactions per hour. The high-speed sample processing is further enhanced by the fact that the Sequence Enrichment Solution utilizes a library of PCR primers in droplets enabling the amplification of hundreds to thousands of genomic loci in a single tube. The RainStorm format avoids the limitations of traditional multiplex hybridization and amplification technologies. RainDance’s solution minimizes process-induced bias or error and requires only a few micrograms of genomic DNA.
